Niacinamide, explained properly.

Niacinamide is one of the most evidence-backed skincare ingredients for the price — but the concentration (2% vs 5% vs 10% vs 20%), the format (serum vs moisturiser), and the co-actives (zinc, peptides, HA) all change what it does. This is the guide that explains all of that, without the marketing.

What niacinamide actually is

Niacinamide is the amide form of vitamin B3 (niacin), a water-soluble vitamin that humans cannot synthesise in meaningful amounts and must obtain from diet (meat, fish, fortified grains) or topical application. In skincare, niacinamide is used at concentrations of 2% to 20% as a leave-on active. It is one of the few ingredients where the oral and topical forms are well-studied independently — the evidence base for each route is separate.

The molecule itself is small, water-soluble, and stable in formulation (unlike L-ascorbic acid, which oxidises rapidly). It does not need a low pH to penetrate the skin, and it is well-tolerated by virtually all skin types. The Cosmetic Ingredient Review Expert Panel concluded in 2019 that niacinamide is "safe as currently used in cosmetics" at concentrations up to 20% — one of the cleanest safety records in skincare.

The bottom line: niacinamide is the safest, most stable, most versatile active in skincare. It works at multiple concentrations and stacks with every other active. The "right" concentration depends on what you're trying to fix: 2–5% for barrier support, 10% for hyperpigmentation and sebum regulation, 15–20% for the rare cases where 10% is not enough.

How niacinamide works (in plain language)

Niacinamide is a precursor to NAD+ (nicotinamide adenine dinucleotide) and NADP+, two coenzymes that are central to cellular energy production, DNA repair, and redox reactions. Topical application raises the local concentration of NAD+ in the skin, which has measurable effects on several cell processes:

  1. Inhibits melanosome transfer. Niacinamide blocks the transfer of melanin-filled melanosomes from melanocytes to keratinocytes. Less melanin reaches the surface = less visible hyperpigmentation. This is the mechanism behind Draelos 2006 and Bissett 2009.
  2. Reduces sebum production. A 2006 RCT (Draelos) showed 2% niacinamide reduced facial sebum excretion rate by ~30% over 4 weeks. The mechanism is not fully understood but appears to involve modulation of sebocyte differentiation.
  3. Supports the skin barrier. Niacinamide upregulates synthesis of ceramides and other stratum corneum lipids. A 2000 study (Tanno) showed 2% niacinamide increased ceramide synthesis by ~30% and reduced transepidermal water loss.
  4. Anti-inflammatory. Niacinamide inhibits NF-κB signalling and reduces the release of pro-inflammatory cytokines. Visible result: less post-acne redness, calmer skin, fewer inflammatory breakouts.
  5. DNA repair support. Niacinamide is a substrate for PARP (poly-ADP-ribose polymerase), the enzyme that repairs UV-induced DNA damage. Topical application after UV exposure accelerates DNA repair, reducing the long-term carcinogenic effect.

Visible results timeline: 2–4 weeks for sebum regulation; 4–8 weeks for barrier improvements; 8–12 weeks for visible brightening and hyperpigmentation fading. The 4-week quitting window is too short for this ingredient.

Why concentration matters

Niacinamide is one of the few actives where the dose-response relationship has been carefully mapped. Here is what each concentration range does, based on the published RCT base:

2% niacinamide

The "barrier support" concentration. Modest sebum reduction (~20% per Draelos 2006), measurable ceramide synthesis increase, low irritation. The mass-market moisturiser range (CeraVe PM, Olay Regenerist). Slower brightening than 10%.

5% niacinamide

The "entry-level" concentration for visible brightening. Comparable to 4% hydroquinone for melasma at 8 weeks (Navarrete-Solís 2011), with fewer side effects. Well-tolerated on first use. The right starting point for sensitive skin.

10% niacinamide Evidence sweet spot

The most-cited concentration in dermatology literature. Stronger brightening effect, faster sebum reduction, and the typical inclusion dose in clinical trials (Draelos 2006 used 2% but Bissett 2009 and Rolfe 2014 used 5–10% for hyperpigmentation outcomes). The right concentration for most users with hyperpigmentation, oily skin, or post-acne marks.

15–20% niacinamide

The "professional" or "high-strength" range. Published evidence is thinner — most of the strong clinical data is at 5–10%. The CIR 2019 safety review covered up to 20% and found it safe, but flushing risk rises at this range. Generally not necessary for over-the-counter use.

The start protocol

Niacinamide doesn't have a "retinization" period like retinol, but the concentration and frequency still matter. Here is the protocol that maximises results without irritation:

Week 1–2: Once daily, 5% or 10%

Apply 3–4 drops to clean, dry skin, either AM or PM. Most users tolerate niacinamide in the morning, layered under SPF. Wait 60 seconds before the next product. Expect no irritation or tingling — if you feel flushing or tightness, drop to 5%.

Week 3–4: Once or twice daily, full dose

After 2 weeks of tolerability, you can add a second application (AM + PM). Most users tolerate twice daily at 10% without issues. Sebum reduction is usually noticeable by week 3–4.

Week 5–8: Add a co-active if needed

Hyperpigmentation / post-acne marks

What to pair with niacinamide (and what to avoid)

Pairs well

  • Vitamin C (L-ascorbic acid or derivatives) — different mechanisms for brightening
  • Retinol / retinal — niacinamide buffers retinol irritation
  • AHAs / BHAs (glycolic, salicylic) — alternate days or same routine

Three niacinamide myths that need to die

Myth 1: "Niacinamide and vitamin C cancel each other out"

This is the most-repeated niacinamide myth. It originates from a 2008 Cosmetic Dermatology study that showed a transient flush in some users when LAA + niacinamide were applied simultaneously at very high concentrations. In practice, the reaction is uncommon, mild, and self-resolving. The "cancel each other out" claim has no published support. Use them together — different mechanisms, different targets.

Myth 2: "Higher percentage niacinamide is always better"

The dose-response curve for niacinamide flattens around 10%. Higher concentrations (15–20%) appear in some "professional" lines but the additional benefit is marginal while the irritation cost rises. CIR 2019 covered up to 20% and found it safe — but "safe" is not the same as "more effective". Stick with 10% unless you have a specific reason to go higher.

Myth 3: "Niacinamide is just a B vitamin, so you can swallow it instead"

Oral niacinamide (as a supplement) has a separate evidence base for general health, energy metabolism, and DNA repair — but it does NOT reach the skin at concentrations high enough for visible brightening. The skin benefits are only reproducible with topical application. If you want hyperpigmentation fading, you need topical niacinamide, not oral.

What's the difference between niacinamide and niacin?

Both are forms of vitamin B3. Niacin (nicotinic acid) causes flushing at therapeutic doses; niacinamide (nicotinamide) does not. In skincare, only niacinamide is used topically — niacin causes skin irritation and flushing. If a product label says "niacin", it's almost always referring to niacinamide in practice.

Can I use niacinamide with retinol?

Yes — and this is one of the best-supported combinations. Niacinamide buffers retinol irritation (Draelos 2006) and the brightening effect of niacinamide complements the cell-turnover effect of retinol. Many products combine them in a single formula.
